Main focus was product development of a fully automated diagnostic ischemia assay on a Cobas Mira, Plus chemical analyzer. Direct liaison with a clinical chemist and a FDA consultant to facilitate the proper quality assurance of the assay for future clinical trials and FDA 510K Invitro Medical Device approval. This work was contracted by Ischemia Technologies, Inc.

 

Assisted in the planning, organization, and implementation of preclinical trials with heart or severe head trauma patients by processing human blood samples, performing the manual ischemia diagnostic assay, and documenting the results. Liaison with the SMC hospital chemistry department to collect data from DADE Dimension Analyzers for the diagnostic assay and for related biological markers of interest. Carried a 24 hour pager for participation in an Immune Ischemia study.  Prepared and assayed blood samples for various immune markers using techniques such as RNA isolation, ELISA, and fluorescent antibody staining.

In vitro culture of human and mouse coronavirus, titration and purification of live virus, cell culture of various cell lines, administrative duties such as inventory and ordering, edit grants, write letters of correspondence, oversee OSHA documentation and regulations, assume radiation safety tasks and assist/train laboratory personnel. Molecular Biology techniques utilized in project designed to detect coronavirus in human multiple sclerosis autopsy brain included:  preparation of RNA, mRNA and cDNA;  detection of RNA and cDNA with gel electrophoresis; design of PCR primers; isolation of PCR products; bacterial TA-cloning and SequenaseII-sequencing resulting in the observation of mouse-like coronavirus sequences from human brain.  Cell culture and assays involving primary human brain endothelial cells plated on porous membranes for the purpose of measuring migration of virus or immune cells through the confluent cell layer, representing a blood-brain-barrier model.  Preparation and detection of diverse cell types by single or double staining on a Beckman MacIntosh Fluorescent Associated Cell Sorter (FACS).  Blood preparation, cell staining, data aquisition and analysis for FACS for an in-house clinical study.  Set up sorting capabilities on the FACS and became proficient on the machine with minimal training.  Assist FACS users in machine software and hardware operation and maintenance.

Three years experience with molecular biology techniques including:  cloning design for proper orientation and framing using various plasmids; restriction digestion; dephosphorylation cleavage; Klenow fill-in; ligation; transformation; DNA purification from cell culture, agarose, and polyacrylamide gels; radioisotope labeling with 3H, 32P, 35S and 14C; radiolabeled in vivo expression; coupled transcription translation in vitro expression; and DNA sequencing.  Contributed to development of rapid plasmid isolation method.
